Saponin accumulation in the seedling root of Panax notoginseng

چکیده
BACKGROUND Panax notoginseng is an important Chinese medicinal plant. Dammarene-type triterpenoid saponins are main pharmacologically effective compounds in P. notoginseng. This study aims to investigate the formation and accumulation of saponins in P. notoginseng roots during germination and juvenile stage. METHODS P. notoginseng seeds were collected and stored in wet sand. After germination, the seedlings were transplanted into a soil nursery bed and cultivated for one year. During this period, samples were collected every month and the concentrations of ginsengnosides Rg1, Re, Rb1, Rd and notoginsengnoside R1 were determined by HPLC. RESULTS There was little saponin in the P. notoginseng seed. The chemical composition of seed was different from that of root. After germination, Rb1, Rg1, Re, Rd and R1 appeared successively in the seedling root. And in the five-month-old root, all these five main saponins came into existence. The accumulation of saponins in P. notoginseng root was affected by seasons. CONCLUSION The accumulation of saponins showed a time-dependent increase after germination of P. notoginseng.

The present study monitored the effect of 2, 10, and 50 mg/L of Panax notoginseng saponin exposure following hypoxia-reoxygenation injury in fetal rat cortical neurons. Results showed that varying doses of Panax notoginseng saponin significantly enhanced the cell viability of neurons, reduced malondialdehyde content, increased superoxide dismutase activity, inhibited mRNA and protein expression...
متن کاملAutotoxic Ginsenosides in the Rhizosphere Contribute to the Replant Failure of Panax notoginseng
BACKGROUND AND AIMS Sanqi ginseng (Panax notoginseng) growth is often hampered by replant failure. In this study, we aimed to examine the role of autotoxicity in Sanqi replant failures and assess the role of ginsenosides in autotoxicity. METHODS The autotoxicities were measured using seedling emergence bioassays and root cell vigor staining. The ginsenosides in the roots, soils, and root exud...
متن کاملStrengthening Triterpene Saponins Biosynthesis by Over-Expression of Farnesyl Pyrophosphate Synthase Gene and RNA Interference of Cycloartenol Synthase Gene in Panax notoginseng Cells.
To conform to the multiple regulations of triterpene biosynthesis, the gene encoding farnesyl pyrophosphate synthase (FPS) was transformed into Panax notoginseng (P. notoginseng) cells in which RNA interference (RNAi) of the cycloartenol synthase (CAS) gene had been accomplished. Transgenic cell lines showed both higher expression levels of FPS and lower expression levels of CAS compared to the...

Panax notoginseng saponins (PNS) are among the most important compounds extracted from Panax notoginseng root, and have long been used in traditional Chinese medicine to control bleeding. PNS have recently garnered attention for the treatment of circulatory system diseases. The present study aimed to evaluate the effects of PNS on angiogenesis in vitro and to explore the molecular mechanisms un...
متن کاملMolecular Cloning and Characterization of PnbHLH1 Transcription Factor in Panax notoginseng.
Panax notoginseng has been extensively used as a traditional Chinese medicine. In the current study, molecular cloning and characterization of PnbHLH1 transcription factor were explored in Panax notoginseng. The full length of the PnbHLH1 gene obtained by splicing was 1430 bp, encoding 321 amino acids. Prokaryotic expression vector pET-28a-PnbHLH1 was constructed and transferred into the BL21 p...
